I look at the tires and see what is the max pressure at the max weight carrying capacity. If you know the weight of your equipment that you will be carrying and it matches the combined max capacity of the tires it's easy. Just inflate to maximum. Otherwise, I would put the equipment on the trailer first and inflate both tires the same to the point where they have a nice flat tread footprint on the ground. Too much and the center wears faster. Too little and the outsides of the tread wear more quickly.

At home I dont care if my trailer is loaded or unloaded because my compressor is big enough to do the job either way. If I stop at a gas station I will only fill the tires if the trailer is empty because I have found gas station air compressors to be rather weak and slow.

it doesn't matter if the trailer is loaded or empty, what does matter is that you check them before you pull the trailer. Always check them cold.
